Indirect Elisa Procedure. This technique will detect hiv within 5 weeks. See direct elisa protocol buffers and reagents.
The antigen for which the sample must be analyzed is adhered to the wells of the microtiter plate. Method coat microtiter plate wells with 100 µl of the antigen solution at a concentration of between 1 10 µg ml in coating. Wrap coated plates in plastic wrap to seal and incubate for 2 hr at 370c in an incubator.
Indirect elisa detects antibodies from the test sample.
Bovine serum albumin is used to block the other protein binding sites primary sample antibody is added to the plate and incubated with the antigen. Incubate for 1 hour at 37 c. Pipette up and down five times. In tube 1 prepare a 4 ml solution of secondary reagent at 200 ng ml in pbsn.
